As a highly selective PDE5 inhibitor, avanafil competitively antagonizes cyclic guanosine monophosphate, leading to vasodilation of penile blood vessels and enhanced erectile function. Unlike some other PDE inhibitors, avanafil exhibits a high ratio of inhibiting PDE5 compared to other PDE subtypes, allowing for effective treatment of ED while minimizing adverse effects. Avanafil was approved by the US Food and Drug Administration (FDA) for the treatment of erectile dysfunction on April 27, 2012, and subsequently by the European Medicines Agency (EMA) on June 21, 2013.
